What is karyotype analysis?

Karyotype analysis refers to the process of artificially pairing, numbering and grouping the chromosomes of the cell to be tested according to the inherent chromosome morphological and structural characteristics of the organism and according to certain regulations, and performing morphological analysis. The chromosomes of different species have their own specific morphological structure (including the number of chromosomes, length, centromere position, arm ratio, satellite size, etc.), and this morphological feature is relatively stable. The dyed or fluorescently labeled chromosomes can clearly and intuitively observe the specific morphological structure of the chromosomes through certain optical or electrochemical color development equipment, and then compare with the normal karyotype to find differences, and then determine the deletion, duplication and chromosome phenomena such as inversion.

  • Routine morphological analysis.

  • G-banding technology is to color different regions of chromosomes by Giemsa staining method, so that the chromosomes show light and dark bands under a light microscope. Each chromosome has a specific band pattern, and even the long and short arms of each chromosome are specific. According to the different banding types of chromosomes, the individuality of chromosomes can be identified more carefully and accurately.

  • Colored segment analysis.
  • The chromosomes are processed by low temperature, KCl and enzymatic hydrolysis, HCl or HCl and acetic acid mixed liquid, and then processed into slices, which can cause the heteropyknosis reaction of the chromosomes and make the heterochromatin segments visible. The coloring segments are basically the same between homologous chromosomes, but there are differences between non-homologous chromosomes.
